John was born about 1771 based on his age at death (he was in his 75th year), the son of Joseph Jackson and Ann Shute. He married Amy Watson on 29 Mar. 1790 at Trinity Episcopal Church in Swedesboro, Gloucester Co., NJ. They had 11 children together based on his obituary. Ten daughters are named in his estate papers, all linked below. The "missing" child might have been born during the gap in their children's births between 1810 and 1816.

John was a founding trustee of the Union Methodist Episcopal Church in Unionville (Aura), Gloucester Co., NJ, in 1807, when his father granted the congregation 4 acres for its church and burial ground. Under his father's 1810 will (of which he was co-executor), he inherited half of his father's 800+-acre tract at Aura. He and his wife granted portions of this acreage to their sons-in-law, which passed down to descendants.
