Moses is the oldest child of Judge David Mitchell and Martha 'Mattie' Black and grandson of Moses Black, his namesake. When his extended family moved from PA to the Ohio wilderness (1799), Moses was about 17 years old.

On 24 Feb 1808, Moses married Jane Taylor, daughter of Daniel Taylor and Mary Brown, pioneers from VA who settled in the area. They had at least five children: Martha J (Courtright), Daniel, Moses Jr, David and George Mitchell. Unfortunately, Jane died on 20 Apr 1823, a few months after George was born.

About 1826, Moses married Jemima 'Jennie' Cartright or Courtright. They had at least seven children: Malissa D (Snodgrass), Elizabeth 'Betsy' (Taylor), Abraham (died unmarried), Aaron (died unmarried), Amanda F (died unmarried), Margaret A (died unmarried), and Elijah Mitchell.
