Moses was the son of Moses Mitchell and Jane Taylor. He married Mary 'Polly' Marshall about 1836 and then, moved with his family to Iowa about 1850. It is believed that he died before 1860, as he was absent in the 1860 US Federal Census. His father, Moses Mitchell Sr, died around Christmas in 1863. In January 1865, Moses Mitchell Sr's estate was settled in Union County, Ohio. Three of Moses Mitchell Jr's children were underage and represented by guardian, their uncle David Mitchell.
