John Aquilla Cook, Sr. was the son of John Bela Cook and Martha Ann Kelley Cook. On 14 March 1886, John Aquilla Cook, Sr. and Mary Weston were married in Wise County, Texas. In 1889, Mary Weston Cook died. On 23 September 1895, John Aquilla Cook, Sr. and Nettie Barron were married in Angelina County, Texas.

John was the 7th great grandson of Stephen Hopkins of the Mayflower and 6th great grandson of Mayflower passenger Giles Hopkins. Stephen Hopkins was a signer of the Mayflower Compact. Stephen Hopkins was also shipwrecked in Bermuda with John Rolfe on his way to Jamestown, Virginia. In 1610, Stephen Hopkins arrived in Jamestown.

Two of John's ancestors are documented Daughters of the American Revolution soldiers and patriots. These ancestors are Jesse Cook and John Cole.
