The son of the Apostle of the Rappahannock married Clara Forsyth Mason on 10 July 1860 in Fairfax County, Virginia. Their children were Clara Forsyth, Murray Mason and John Peyton McGuire III. John Peyton McGuire II founded McGuire's University School in 1865 to teach a new generation of Southern boys honor, courage and duty, plus skills that guaranteed advanced placement in the University of Virginia. He and his son read the verse from Psalm 15 daily for 78 years to 10,000 boys.
