John Banning, Jr. was the son of John and Elizabeth (Black) Banning. Born in Rockbridge County, Virginia, he moved as a young man to Marshall County, Virginia, settling at Grave Creek (now Moundsville, West Virginia), near Wheeling. He married Eliza Ann Anderson in 1834 and had 10 children by her.

John and Eliza Ann moved west to Henry County, Iowa, about 1838, settling in Baltimore Township on the Skunk River near Mount Pleasant, where John farmed. Eliza Ann died in 1855.

By 1860, John had remarried and had one child, Ephraim Morgan Banning, by his second wife Mary. According to the 1860 Census, two separate "households" are listed, the second of which consisted of John's children by Eliza Ann. (Sons Horatio and John A. Banning were 26 and 24, respectively, in 1860.)
