Son of Martha Elizabeth Howard Murphy and John Henry Murphy, Sr., Civil War veteran and founder of the Afro-American newspaper in Baltimore, MD in 1892.

Married Lulu Vashti Turley in Washington, DC on 20 Jun 1916.
Together, they had the following five daughters: Martha, Ida, Carlita and Vashti (twins), and Frances.

In 1918, he began working at his father's newspaper, and after his father's death in 1922, Carl took over the newspaper.

In 1960, the Carl J. Murphy Fine Arts Center was dedicated in his honor on the campus of Morgan State College, where he served as Chairman of the Board of Trustees. He was also a member of the Board of Trustees for the Baltimore chapter of the NAACP.

Maternal grandfather of the first female bishop of the A.M.E. Church, Vashti Murphy McKenzie.
