Prince Leopold was the first child of Hereditary Prince Frederick of Anhalt-Dessau-Köthen and his wife Princess Antoinette of Saxe-Altenburg. He was named after his grandfather Duke Leopold of Anhalt-Dessau-Köthen. Leopold was married on May 26, 1884 at Schloss Philippsruhe in Hanau to Princess Elisabeth of Hesse-Kassel, the eldest daughter of Prince Frederick William of Hesse-Kassel and Princess Anna of Prussia. They had one daughter, Antoinette, who was married on May 26, 1909 to Prince Frederick of Schaumburg-Lippe.

His remains have seen little rest since the original interment. He was originally interred in the Schlosskirche, Dessau until 1898, at which time he was reinterred in the Mausoleum, Dessau. The remains were then reburied in an unmarked grave in the Friedhof Ziebigk, Dessau, following World War II. Finally, he was reburied in 2019 in the Marienkiche, Dessau.
