Henri I, Duc de Guise, was the eldest son of François de Lorraine, Duc de Guise, and his wife, Anna d'Este, the eldest daughter of Ercole II, Duke of Ferrara (son of Lucrezia Borgia and grandson of Pope Alexander VI) and his wife, Renée of France (daughter of King Louis XII of France) . His maternal grandparents were Ercole II d'Este, Duke of Ferrara, and Renée of France, daughter of King Louis XII of France.

He founded the Catholic League in 1576 to prevent Henri, King of Navarre, head of the Huguenot movement, from succeeding to the French throne. A key figure in the French Wars of Religion, he was one of the namesakes of the War of the Three Henrys. A powerful opponent of the queen mother, Catherine de' Medici, he was assassinated by the bodyguards of her son, King Henry III.

He was married on October 4, 1570 in Paris to Catherine de Clèves, Comtesse d'Eu, by whom he had fourteen children.

His body was burned and the ashes thrown into the Loire following his assassination. The magnificent monument his widow erected for him is a cenotaph.
