Muster roll entries state David was 6'1" tall, light hair, blue or brown eyes, with light complexion, and an unmarried farmer. He was residing in Sumner township, Warren county, Illinois when he enlisted on 1 May 1861 at Young America (now Kirkwood), Warren county, Illinois, for a 3 year term. He was mustered into Co. C of the 36th Illinois Infantry at Aurora, Illinois on 23 September 1861. He re-enlisted at Blain's Crossroads near Chattanooga, Tennessee on 25 January 1864 as a veteran. He was mustered out on 8 October 1865 in New Orleans, Louisiana at the rank of Sergeant, although his pension papers list his rank as Corporal. His headstone application gave his rank as Sergeant.

David was joined in Company C by his brother John Russell Henderson in 1862, and brother Robert Alexander Henderson in 1864; all were discharged at the end of the war. A John F. Henderson enlisted with David, listing he was born in Westmoreland county, PA, and dying of wounds in 1863 - a cousin, perhaps.

It would appear that David followed his brothers from Illinois through Iowa to Kansas, then back to Chicago, Illinois. David and his wife operated a crockery store in Chicago. He had retired from business several years before his death due to poor health, and had moved to Denver about a year before his death hoping for an improvement in his health. He was survived by his wife, Margaret, and preceded by his daughter, Cora Lee.
