Florentine Lake was born near Oxford in Coshocton county, Ohio. He was the son of Joseph Lake and Epheline Bedell Lake and had a brother and a sister. When he was two years old his father died of cholera while floating merchandise down the Erie Canal. His mother remarried within a short time to Malcolm Warden and they had two more boys and a girl. In about 1844, at the age of twelve, Florentine left home and traveled by himself to his uncle Cornelius' place in Indiana. By 1850, his parents had also moved to Indiana to join Cornelius and another uncle, Henry Bedell. The 1850 US census shows that Florentine is living with his uncle Henry's wife's father, Philip Lash.

Uncle Cornelius moved again to Linn county, Iowa, and Florentine joined him in about 1852. It was there that he met and married Delilah Scott, daughter of James Scott and Mary Polly Wisehart Scott in 1856. They moved by covered wagon with their first three children to Hardin county, Iowa in 1860. They lived in that county for the rest of their lives.
