James Neville Jr. was the son of Private James Neville, who fought in Virginia and Kentucky during the Revolutionary War. His Uncle was Brigadier General John Neville, who is buried near Pittsburgh. He had two other uncles (Joseph and William) who were Officers in the Revolutionary army. His Aunt Nancy married an officer who fought in the Revolutionary War, as well. He was brought to Lincoln County, Kentucky in 1779 by his parents, and then they moved to Barren County by 1800. They took care of the three grandchildren (Amanda, Rebecca and James) of their son James III, when the first wife (Virginia Bailey) died. Eventually, that son, James Neville III was shot to death in 1863 in Missouri. Additional children included Thomas, Elizabeth (Betsy), Sarah (Sally), Dr. George Washington Neville, John Thomas, and William Thomas.
