Jacob was born in the village of Moos near Buehl in Baden (later part of Germany) to parents Johannes Jakob Kloepfer and Katherina Krummholz. Jacob came to North America on the ship 'Ilzaide' from Le Havre landing in New York on 13 June 1840. His future wife, Monika Winter, was also from Moos and came to North America on the same ship. Jacob and Monika were married in the St. Louis Catholic Church in Buffalo, New York on 22 July 1840.
