John was the son of Job and Mercy (Perry) Caswell. According to the Greene town history, he had two brothers and five sisters. His father and older 14-year-old brother Job both served in the Revolutionary War. His parents and some other family are buried in the Plains Cemetery in Leeds.

John married Christiana Perry on November 5, 1791 in Situate, Massachusetts. He remarried second to Mary Jepson on September 27, 1815. John had large families by both of his wives. Mary was from China, Maine (formerly called Harlem),and according to the New England Historical and Genealogical Register, they were married in Harlem.
