Samuel was born to Joseph Peden and Mary Mills in Antrim Co. Ireland. His father gave him permission to serve as an apprentice to a ship captain, who then sold him to Robert Smith a market gardner at age 13 or 14 after the ships arrival in Baltimore in about 1743-44. After he had served his apprenticeship, he was given a horse, saddle and cash He rode from Baltimore to York Co. PA and later met and married Lydia Porter in 1751.

They settled in Lancaster Co. PA, where all six of their children were born, Obediah, Samuel, Lydia, Joseph, Isaac and Alexander Peden. Later they moved to WA Co. PA.

Samuel and his wife were active members in the Quaker Church.
