Samuel was the firstborn son of Samuel S. Jeffries and Nellie May Anderson Jeffries. He grew up at 2146 Ellsworth Street in Philadelphia, with his sisters Eleanor, Margaret, Elizabeth and Elsie, and his brother, Clifford. Samuel was probably named for his father, with his mother's maiden name as a middle name, although he could have been named in honor of his mother's uncle, Samuel Anderson. Brother Clifford was named for Nellie's brother, her children's uncle Clifford Anderson, and sister Elsie may have been named for their aunt, Elsie Anderson Thompson. It is unknown to this researcher if Eleanor was named for Nellie's grandmother, Eleanora Seal Anderson.

Sam's parents instilled a strong sense of 'family' into their youngsters, as they graciously took in their elderly parents, Mary Jeffries, mother of Samuel S. Jeffries, and Isaac Anderson, father of Nellie May Anderson Jeffries, and at times other relatives in need.

Both of Sam's paternal great grandfathers, the fathers of his grandparents, Isaac and Mary A. Jeffries, hailed from England. His maternal great grandfather, John Anderson, father of grandfather Isaac Anderson, hailed from Scotland.

Samuel Anderson Jeffries' father, Samuel S. Jeffries, was a Teamster, and young Sam began working as a shipper when he was a teenager. He married Mary Jane Gamble in Philadelphia in 1923, and they had five children. Following his parents' example, Sam and Mary also took in relatives in need.

Samuel and Mary began their family in Gloucester, New Jersey, later settling in Upper Darby, Pennsylvania. Sam worked as a sales agent for electronics/radio parts. His wife Mary's parents were born in Northern Ireland.
