James Murdoch was the son of Hamilton Murdock, born 1731 in Killyleah, County Down, Northern Ireland, and Mary Cleland born in Killyleah, County Down, Northern Ireland. He was the grandson of William Murdock II, and Martha Hamilton and great grandson of William Murdock I and Jennett Murkland.

James Murdoch was born May 6, 1766. He was two years old and not listed as a passenger on ship register. James Murdock married Elizabeth (Betsy) Davenport in 1791 when he was 25 and she was 16. James and Betsy had been members of the First Creek Baptist Church since 1835. Betsy was born in 1775, the daughter of Isaac Davenport and his first wife, Martha Anne Ridgill Davenport.

Elizabeth (Betsy) Davenport Murdoch's father, Isaac Davenport, was born in 1752, probably in Granville County, NC. He died June 26, 1816. His tombstone is in the Davenport burying ground, ten miles west of the Newberry Courthouse. Isaac was the son of Francis Davenport, who furnished goods and paid for them for the American Revolution.

James Murdock died June 23, 1857 and Betsy died two months later. Markers have been placed on their graves in the First Creek Baptist Cemetery in Abbeville County by Strother E. Murdoch, Sr. and Jr. James and Betsy had been members of the First Creek Baptist Church since 1835.

James and Betsy Murdock had 15 children, whose birth dates are all listed in the Murdock family Bible. All the children were born in Newberry, SC.
