Luise was born the daughter of Frederick Wilhelm III, King of Prussia and his consort, Luise Augusta, Princess of Mecklenberg-Strelitz in Berlin, Brandenburg, Germany.

On 21 May 1825, she married Willem Frederick Karel, Prince of the Netherlands in Berlin.

They were the parents of 4 children-Wilhelmina Fredericke Alexandrine Anna (wife of King Karl XV of Sweden), Willem Frederick Nicholas Karel, Willem Frederick Nicholas Albrecht, and Wilhelmina Fredericke Alexandrine Anna Luise Maria.
