Manley Cornelius was the son of Beverly Cornelius and Nancy Euphemia Smith and grandson of William and Lettice Cornelius. He was married to Martha Hooper, daughter of John Hooper, on September 25, 1845 over in St. Clair County. They had four children: Thomas Benton Cornelius, Indiana Cornelius (died young), Nancy Hester Cornelius Sims, and Rachel C. Cornelius Pearson (died after birth of first child).

Manley served in the 28th Alabama Infantry Regiment during the Civil War. It is said he gave the wood for the sub floor in the old Clear Springs Methodist Church sanctuary.
