John Cardwell was the son of Thomas Cardwell and Martha (Perrin) Cardwell, born in Henrico County in 1715.

He married Keziah Lowe in 1742 in Goochland County, Virginia.

Their marriage produced twelve children or more, all born in Virginia, including:
Thomas Perrin, 1745-1828; Ann, 1747-1818; Elizabeth Nancy, 1747-1795; John Jr, Abt 1748-Aft 1825; Mary, 1749-1795; Perrin, 1752-1854; William, 1756-1796; Martha, 1758-1838; James, 1761-1834; Lucy, Abt 1766-Abt 1795; George, 1769-1834.
