Born to Benjamin and Lucinda (Newell) Tilton. His father was the founder of the Harvard Bank (later known as the First National Bank of Cambridge and later the Harvard Trust Co.)

He graduated from Harvard with an A.B. in 1862 and A.M. in 1865. He married Ellen Trowbridge July 21, 1864. They had three sons; William, Benjamin and Newell, and one daughter, Ellen Maud Tilton Atherton.

He spent many years in education. In 1864 he taught for three years at Highland Military Academy in Worcester, MA. In 1867 he served as the superintendent of public schools in Newport, RI. In 1871 he became principal of Phillips Academy in Andover, MA. From 1873 to 1890 he was head master of Rogers High School in Newport, RI. He moved to Cambridge, MA in 1894 and entered into the banking businesses established by his father.

He was President of the Cambridgeport Savings Bank and director of the Harvard Trust Co. He died suddenly after dinner with friends at the Cambridge Club.
