Mary Cotton Vance was a daughter of Henry and Mary Cotton of Bedford County, PA. Her siblings were: Samuel, Hugh, Rachel, John, Lydia, Martha and William.

She married Isaac Vance on 15 November 1783. They lived on their farm in South Strabane where the following children were born: John, Henry, Isabell, Samuel, Mary, Martha and Isaac. Seven other children died early in life.

Mary and Isaac were among the founders of the Pigeon Creek Presbyterian church and both are buried there.
