Acuff Chapel was the first Methodist church established west of the Appalachian Mountains in what later became the state of Tennessee. It was built in 1786 and is located on highway 126 near Blountville. His wife was Anna Acuff (1742-1820).

His mother was Sarah Winn Ascough, born in 1698 in Virginia. He had an older brother, Christopher Ascough, Jr who was born aft 1721 and starting with Timothy and on down with his other siblings they had changed the name to acuff for some reason. His other siblings were Jane Acuff (she married Joseph "Jobe" Breeden), John Acuff, Sr and Cain Acuff.
