John Antle was a son of Henry and Mary Antle. He was married in Kentucky to Elizabeth Buchanan, and they had five children born there: Polly, Sally, Henry, Andrew J., and Martha J.

The John Antle family moved to Morgan Co., IL in 1829 and to Salisbury, Sangamon Co., IL in 1830. Rev. John Antle preached to five churches known as Separate Baptists. The churches were located in Salisbury, McKinnie Settlement in Sangamon County, Baker's Prairie, Sand Ridge in Menard County, and one in Morgan Co., IL. He also worked as a farmer, wagonmaker, and blacksmith.
