Jeptha B. Mathis was born the son of Lewis Mathis (1775 – 1834) and Luranah Brantley Mathis (1779 – 1849). He married Sarah L. Davis 28 April 1831 and they had at least five children, Eliza Jane Mathis (1833 – 1855), John L. Mathis (1835 – 1852), Susan Ann Mathis (1840 – ), Sarah E. Mathis (1842 – ), and Mary Lou Mathis (1845 – 1921).
