From a family of physicians. He was the son, grandson, father and grandfather of physicians; He was a research chemist who wrote about malaria, cholera, plague and yellow fever.

He married Sarah Matilda Henry in 1822. They had nine children.

1). Alexander Henry Mitchell (May 12, 1823-May 10, 1839). Originally buried in the Second Presbyterian Burial Ground, Philadelphia.

2). Elizabeth Kearsley Mitchell (January 14, 1824-May 19, 1874).

3). Silas Weir Mitchell (February 15, 1829-January 4, 1914). First married to Mary Middleton Elwyn (d.September 1862), second married to Mary Cadwalader (1835-1915). He was a world famous medical pioneer and physician.

4). John Henry Mitchell (September 18, 1837-April 25, 1846).

5). Sarah Matilda Mitchell Neilson (March 2, 1831-December 12, 1906). Married William Neilson in 1848. Buried in Burlington, NJ.

6). Letitia Henry Mitchell Harrison (August 11, 1832-December 29, 1914). Married George H. Harrison in 1856.

7). Robert Walsh Mitchell (April 25, 1836-April 2, 1872). Captain, 6th Pennsylvania Volunteer Cavalry, Civil War. Died in St. Kitts, West Indies.

8). Nathanial Chapman Mitchell (December 31, 1840-April 26. 1900). 1sdt Lieutenant, 15th Illinois Volunteer Cavalry and Lieutenant Colonel, 4th United States Colored Cavalry, Civil War.

9). Edward Donaldson Mitchell (November 1843-May 31, 1864). Medical Cadet, US Army, Civil War.
