Jacob was the son of Jacob Seek Sr. and Louisa Virginia 'Jennie' Robinson. His siblings were Mary Jane, Malissa Elizabeth, Emma, James, Joseph, Eliza, Sarah Caroline, George, Laura, Ella, Virginia and Allie.

He married Betty Elizabeth Pardue on January 3, 1878 in Ray, Missouri. Their children were Maggie Louella, Minnie, Jacob Frank, Cora, Nellie and Ella.

Paternal grandparents were Joseph Matthew Seek and Mary C. King.

Maternal grandfather was James Robinson.
