Wiley Estes Pinkerton was born on April 12, 1816 in Nashville, Davidson, Tennessee. He was the son of David and Celia Barnes Pinkerton. Wiley moved to Greene County in 1823 and located near Berdan, where he purchased a tract of land on which he resided until his death at only 33 years of age. Following in his father' David's footsteps, Wiley was also a tax assessor and collector for Greene County.

He married Sarah P. Allen on October 19, 1843, the daughter of John Allen and Martha "Patsy" Barrow. In 1844 he bought 34.68 acres next to his father's property for $43.35. On December 1, 1845 he bought (Section 30, Township 11-N, Meridian 11-W, Green county). He and Sarah had two children, Revel Estes and Melissa Jane. His children were only three and one year old when he died.

In 1846 he was appointed guardian for William Jackson Allen, Alonzo Allen, and Nancy Allen younger siblings of his wife Sarah, after their father John Allen, Jr. died.
