Son of Amos Jackson and Mary Ann (Farmer) Cook, Rufus was married 4 Aug 1923 in St Louis to Thelma Flora Irene Van Meter. They had children: June LaFay Lindamood (1924-2009), Margie Lucille (1926-1991), Mosie E. (1928-1988) and John Duglus (1931-1934). They divorced, and Rufus married secondly to Dorothy Lee Tweedy, daughter of James J. and Martha Isabelle (Morgan) Tweedy of Union County, Illinois. Years later they learned that they were distant cousins. Rufus' great grandfather, Frederick Cook, was Dorothy's great great grandfather.

Rufus and Dorothy had 12 children together, of whom 2 are still living.
