Samuel Barker was born in England, near Stoke-on-Trent. His parents were Richard Barker and Ruth (Allerton) Barker. Samuel, his parents, and 3 of his brothers immigrated to America around the year 1750. His father was a sea captain and they originally settled on Martha's Vinyard or Nantucket Island before moving to western New Jersey. Samuel joined the fight during the Revolutionary War on the side of the Continentals, eventually contracting a sickness that caused him to return home for care. He died of this sickness in 1777, leaving behind his wife: Mary (Fithian) Barker, and young son: John Barker.
