John Randolph, along with his wife, actress Sarah Cunningham, were co-founders of the Ensemble Studio Theater in New York City in 1971; they also launched its sister company in Los Angeles in 1980, and they appeared in many of its productions as performers, sometimes acting together.

He and his wife's acting careers were seriously affected by the House of Un-American Activities Committee's (HUAC) about Communism in the entertainment industry, as they were blacklisted from working in Hollywood films and in New York film and television and radio after 1948. In 1955, when they were called to testify, they both refused to answer any questions posed to them, as they both supported themselves by acting estensively in theatre until the late 1960s, when they both began finding extensive TV and film work again.
