Eldest son and heir of Henry Somerset, 2nd Earl of Worcester, and Elizabeth Browne. Paternal grandson of Charles Somerset, 1st Earl of Worcester, and Lady Elizabeth Herbert, 3rd Baroness Herbert. Maternal grandson of Sir Anthony Browne and Lady Lucy Neville.

He married the Honourable Christian North circa 1550. She was the daughter of Edward North, 1st Baron North, and Alice Squire. They were parents to three surviving children.

William was a supporter of Lady Jane Grey in 1553.

After his wife's death in 1563, he married Theophila Newton, daughter of Sir John Newton of East Harptree, Somerset, and Margaret Poyntz. His second marriage was childless.

He died at his house in Hackney, Middlesex, and was buried at Raglan. His widow later remarried to William Paratt of Pantglas.

William was succeeded by his only son Edward.
