Son of Theophilius Howard, 2nd Earl of Suffolk, and Lady Elizabeth Home. Paternal grandson of Thomas Howard, 1st Earl of Suffolk, and Katherine Knyvett. Maternal grandson of George Home, 1st and last Earl of Dunbar, and Elizabeth Gordon.

James married three times. Firstly, on 1 December 1640, to Lady Susanna Rich, daughter of Henry Rich, 1st Earl of Holland. They had one surviving daughter named Essex, and Susanna died on 15 May 1649.

He married secondly, circa February 1650, to Barbara Villiers, daughter of Sir Edward Villiers. She was twice widowed at the time of their marriage. They also had a surviving daughter, Elizabeth. Barbara died on 13 December 1681.

Before 8 May 1682, James married as his third wife, Lady Anne Montagu, eldest daughter of Robert Montagu, 3rd Earl of Manchester, but their marriage was childless.

He was succeeded by his younger brother George in the earldom of Suffolk, while the barony of Howard de Walden fell into abeyance between his two daughters for nearly a century, until it was called out of abeyance for a descendant of his elder daughter Lady Essex Howard, Baroness Griffin.

The title has since passed through his descendants by his younger daughter Lady Elizabeth (Howard) Felton, most recently being called out of abeyance in 2004 in favour of Mary Hazel Caridwen Czernin, eldest daughter of the 9th Baron.

The 3rd Earl of Suffolk is buried in the Howard Vault. The vault, now completely bricked in, was closed in 1860.
