Daughter of Thomas Howard, 1st Earl of Suffolk, and Katherine Knyvett. Paternal granddaughter of Thomas Howard, 4th Duke of Norfolk, and the Honourable Margaret Audley. Maternal granddaughter of Sir Henry Knyvett and Elizabeth Stumpe.

Her siblings included the 2nd Earl of Suffolk, the 1st Earl of Berkshire, the 1st Lord Howard of Escrick, the Countess of Salisbury and the Countess of Somerset.

Elizabeth was first married to William Knollys, 1st Earl of Banbury, who was 39 years her senior. Though two sons were born during their marriage, they have long been believed to be fathered by Edward Vaux, 4th Baron Vaux of Harrowden.

Less than five weeks after the death of William Knollys, Elizabeth married Lord Vaux.

Their marriage remained childless however, and without a legitimate son, the barony of Vaux devolved upon the 4th Baron's younger brother Henry Vaux.

Elizabeth is buried in Dorking, Surrey.
