Son of Jacob Crouse (b. 1769 in Montgomery county, NY) he was of German ancestry, his grandfather George having been born in Palatine Saxony in 1740. John began his working career as a store clerk and in a few years formed a partnership with his brother Daniel in a general merchandise business in Canastota. After 28 years the partnership dissolved and he came to Syracuse and established a wholesale grocery store with his brother James. He also became involved in banking and was one of the first directors of the first National Bank. On June 16, 1831 he married Catherine, daughter of Ansel White of Whitestown, NY. She died in 1885 and it was in her memory that he erected the "John Crouse Memorial College for Women" at Syracuse University. He also contributed to The Home, The House of the Good Shepherd and the Hospital.

"Mr. Crouse was undoubtedly the richest man in Syracuse. His wealth consists principally of ready money, government bonds, and railroad stocks which paid dividends. Mr. Crouse was one of the largest holders of New York Central & Western Union Telegraph stock. His real estate amounted to an exceedingly small fraction of his fortune. A gentleman, who was as near the family as any body could be, said last night that D. Edgar Crouse, who will inherit the wealth of his father, will be worth $10,000,000."
