Isaac is the son of Isaac Puffer and Jane McQuay. His first wife was Josephine Mary Weeks, passed away in 1899. They had three children, Ina Villet, Asa Ernest, and Candace. In 1901 Isaac married Bessie Weeks, Josephine's sister, at a Methodist Church in Iowa. I.M. Puffer arrived to Miami late in life where he is buried at Woodlawn Park North Cemetery. His occupation was a Methodist Minister.

Isaac and Emma's three children are Emma Oma, Ina Willetta and Kenneth. Children are all buried in other states.
