Isaac Lovejoy was a son of Samuel Lovejoy (1693-1749) and Hannah Stevens Lovejoy (1692-1767). His grandparents were Lieutenant William Lovejoy (1656-1748), Mary Farnum Lovejoy (1666-1739), John Stevens, and Ruth Poor Stevens.

Isaac married Deborah Sheldon (1723-1813) in Massachusetts on Feb. 28, 1750; she was a daughter of Samuel Sheldon and Sarah Hutchinson Sheldon. Isaac's and Deborah's children included Deborah Lovejoy Ordway, Samuel Lovejoy, Abigail Lovejoy Morse, Sarah Lovejoy Hubbard, Isaac Lovejoy, Jr., William Lovejoy, Persis Lovejoy, Lemuel Lovejoy, and Lydia Lovejoy Downing.

Lovejoy sons Samuel and Isaac, Jr. married daughters of William Morse and Phebe Bodwell Morse; Samuel married Esther and Isaac, Jr. married Mary ‘Polly'.

"Isaac Lovejoy served under Captain Joseph Butler, 1st Co. Colonel John Nixon, 5th Regiment, Massachusetts Soldiers, American Revolution. A receipt for advanced pay was signed by Isaac Lovejoy, June 10, 1775. Private in same comapny (sic) and regiment, RollCall, Aug. 1, 1775. Service 2 months and 8 days. Received order for bounty coat or its equivalent in money, Dec. 20, 1775."
