In the late 1950's, he organized the successful senatorial and Presidential campaigns for John F. Kennedy. He served as the Congressional liaison representative for Presidents Kennedy and Lyndon B. Johnson, and had a varied career in politics, government and business. In 1965, President Lyndon Johnson appointed him the 57th Postmaster General. Serving until 1968, while in office he put into motion a plan to modernize the postal system, making it a quasi-private corporation. O'Brien was twice named chairman of the Democratic National Committee and was chairman the committee's offices in the Watergate office, when the complex in Washington was burglarized, resulting in scandals that led to President Richard M. Nixon's resignation. He also served as the Commissioner of the National Basketball Association (1975-84). He died at age 73 while on business in New York City.
