James Moore's family was part of the many hundreds of Scots Presbyterians who settled in the northern counties of Ireland, becoming known as the Scots Irish. Likewise, hundreds if not thousands left Ireland in the early part of the 18th century, to settle in North America.

James married Jane Walker in Chester County, Pennsylvania, in April 1734. Jane was the daughter of John Walker and his wife Katherine Rutherford, also Scots Irish Presbyterians, from Wigton, Scotland, by way of County Down, Ireland, who immigrated to Chester County, Pennsylvania (some researchers state Maryland) about 1730.

About 1736, James, along with his father-in-law John Walker and some others, scouted out new lands to settle in the Valley of Virginia. In 1743 he was granted 327 in the Borden Tract.
This property was located on Walker's Creek east of Jump Mountain, in today's Rockbridge County, Virginia.

In their later years, James and Jane made their home with their daughter Rachel McPheeters on the Middle River in Augusta County.
