Samuel Boruff was the son of Valentine Boruff & Margaret Cote/Mallicote/Mallicoat Boruff. He married Elizabeth Butcher daughter of Barnaby Butcher and Margaret Mosier on 15 July 1817 in Tazwell, Claiborne County, Tennessee.

They had sixteen children: Mary (Siscoe), Anna (twin of Mary), Sarah Ann (Carter), Henry Valentine, Connie (Siscoe), Samuel Franklin, William Henry, Solomon Butcher, Eli James K. Polk, Green Lewis, Elizabeth E. (Anderson), Margaret Jane (Campbell), John Perry, Thomas Jefferson, David Marion, and Daniel Washington Boruff.

After Samuel's first wife's death he remarried to Eleanor Jones O'dell on 28 December 1860 in Monroe County, Indiana. They had one son, Dougan Boruff.
