Current research puts much into question regarding James' origins. Several family trees suggest he was not the original immigrant of the line, but either the son or grandson of the first Moore immigrant of the line. Additionally, there are questions as to whether the line was of English, Scots, Scots-Irish (Ulster), or Irish (Cork) origin. There is nothing known that states origin of the line. Analysis of the Y chromosome for Moore men directly descended from James Moore reflect a terminal SNP of BY3368.

Other notes of interest:

DNA is suggesting James Moore may have been a brother to Ruth Moore, wife of Col. Ninian Beall.

A daughter, Mary, married John Summers (ca. 1685-1769).

Alexander Moore (b. 1777, Maryland, and died in Kentucky) is, according to DNA, not a Y descendant of James. Male descendants of Alexander have been shown with haplogroup I-M253 (see Group 69 of the Moore DNA Project, kits 304902 and 57760). Male descendants of James Moore are from haplogroup M269.That being said, however, Alexander was not the son of a Moore, but the daughter of one who was descended from James Moore, and subsequently carried the Moore name, nonetheless.

It has also been learned, via DNA, that James Hook (born ca. 1680-1688) was not the son of Thomas Hooke/Hook and Annaple Beall, but a son of James Moore and Annaple Beall. Y DNA and autosomal DNA triangulations have been performed to confirm this.
