James Madison Riley was a son of James Taylor Riley and his second wife, Martha "Patsy" Cass/Cast. He had nine full siblings. He also had two older half-sisters from his father's first marriage to Susan Lanneau/Lanham and a younger half-brother from his father's third marriage to Susan Mockabee/Mockerbee.

James Madison Riley married Adeline Josephine Benoist on September 20, 1832 in Clay County, Missouri. Another possible marriage date is April 22, 1835 in St. Louis, St. Louis, Missouri. They had, at least, four children.

A death notice, published November 2, 1883, in the Liberty Tribune of Clay County, Missouri, indicated James Madison Riley died "in St. Louis last Fri."

The death notice mentions his uncle, James T. Riley and his brothers, Alfred M. Riley and William Riley. William Riley was actually his younger half-brother from his father's third marriage to Susan Mockabee/Mockerbee.

The death notice also mentions James Madison Riley moved to St. Louis about 1849 from Clay County, Missouri.

James Madison Riley was 73 years old at the time of his death.
