Joseph was born in Benteler, Westphalia, Prussia (modern-day Germany) to Johann Heinrich Plassmeyer (née Nienkemper) and Maria Elisabeth Heckemeyer-Plassmeyer. He immigrated to the United States with his family in 1857.

Joseph enlisted in Company D of the 26th Missouri Volunteer Infantry on Sept. 20, 1861, at St. Aubert, Missouri. He was promoted to 5th sergeant before he was hospitalized and honorably discharged on Dec. 16, 1862. His brothers Anton and John died in the war.

Joseph was married to Margaretha Bode-Winkelmann on Feb. 6, 1867, at St. Joseph Catholic Church in Westphalia, Missouri, and they had five children. Margaretha also had two children from her previous marriage to Johann Gerhard Winkelmann. After her death, Joseph was married to Elizabeth Morfeld on May 1, 1883, at St. Joseph Catholic Church, and they had a son, Joseph.

Joseph was the director of the Bank of Westphalia as well as a farmer.

Joseph died of an epithelioma (skin cancer) when he was 89 years old.

A grand and good man yielded up his life when death called Joseph Plassmeyer home last Friday. Mr. Plassmeyer was 89 years old on the 7th of March. Mr. Plassmeyer was married twice, his first wife being Miss Margaret Winkelmann, of which union were born the following children: Rev'd Henry Plassmeyer, Martin Plassmeyer, Mrs. Henry Adrian, and Mrs. Schwartze, the wife of Judge B.F. Schwartze.

Some time after his wife's death, Mr. Plassmeyer was united in matrimony with Miss Elizabeth Morfeld of which marriage Major Joseph Plassmeyer of the Regular Army was born. Mr. Plassmeyer's funeral was held at the Catholic Church at Westphalia at 8 a.m. Monday, Rev. Henry Plassmeyer officiating, assisted by Rev. John C. Melies, pastor of the Church, and Rev. Joseph Winkelmann, pastor at the Rich Fountain parish.
