Job grew up in Plymouth and worked as machinist before becoming an undertaker for the City of Boston. He lived in Andover before moving to South Boston. While in Andover, Job went into business with Ephraim Everson, and their business was known as Everson & Cole. Their co-partnership dissolved in October of 1841, and at that time Job took over the business. While in South Boston, he worked with his nephew Jabez "J.B." Burgess Cole who took over the undertaking business upon Job's death. The business was renamed "J.B. Cole & Son" with J.B.'s son Harry Holbrook Cole working with him.
Job's official cause of death was from Bright's Disease of Kidneys.

Funeral services over the remains of Job T. Cole were held this afternoon at his late residence, 165 Broadway. Rev. L. V. Price of the E Street Congregational Church officiated, assisted by Rev. John Wright, rector of St. Matthew's Episcopal Church on Broadway. The house was crowded by sorrowing friends of the deceased, and there were a large number of floral tributes sent in. The body will be taken on the 8 o'clock train. Old Colony railroad, tomorrow morning for interment in Plymouth." - Boston Daily Globe, August 10, 1883.
