John was a son of Elizabeth Case and John Barber (also spelled it Barbour). His siblings were: Elizabeth, Rhoda, Cynthia, Abi, Sylvia (died within one year), Sylvia, Luke and Austin.

"John Barbour was a Deacon. He came to Johnsonburg, New York from Connecticut and took up land on Centerline Road near the top of Barbour Hill directly west of Johnsonburg. It is assumed that he emigrated here with his relatives from Connecticut, as they all settled and purchased adjoining lots on Centerline Road in Johnsonburg, New York. Jared bought Lot 20 in 1811 and also 1814; Roswell (Jared's brother) bought Lots 13 and 21 in 1807 and part of Lot 12 in 1811. John established the Barbour farm on top of Barbour Hill between Johnsonburg and Humphrey's Hollow, New York. "

He first married Delight Griswold Case October 13, 1803 and had 3 children: Lucius A., Evelina G. and Edwin Case. Delight died in 1811.

John then married Fanny Hunt on June 15, 1812. They had 10 children: Seldon, Fanny Maria, Fedelia Gates, Herschell, Theodore Dwight, Silvia, Goodrich Hollister, John Newton, Theron LaSalle and Juliet Louise.

John was the first one in his branch of the family to change his last name from Barber to Barbour.
