Lecturer of philosophy at the Ambrosian Seminaries of Milan, and later of theology at the renowned Catholic University of the same City, Maggiolini served furthermore as diocesan assistant of the FUCI, episcopal vicar for the University of Milan, diocesan assistant for Catholic jurists, and director of a cultural review regarding the Italian Catholic Church.
Aged 51, he was appointed by Pope John Paul II, as Bishop of the Diocese of Carpi following the retirement of Msgr. Artemio Prati (1907 – 2004), receiving his episcopal consecration on May 29, 1983, from Cardinal Sebastiano Baggio, assisted by the named Bishop Prati and Gilberto Baroni.
Following the retirement of Msgr. Teresio Ferraroni (1913 – 2007), Maggiolini was named as his successor as Bishop of the Diocese of Como, on January 31, 1989.
During his eighteen years of episcopate in Como, Msgr. Alessandro formed part of the Episcopal Commissions for Doctrine, Faith and Catechism, and for Culture and Schools, and consecrated to the episcopate Bishops Franco Festorazzi, Dante Lafranconi and Oscar Cantoni.
Retiring from the pastoral government of his see at the canonical age of 75, on December 2, 2006, he was succeeded by Msgr. Diego Coletti.
A renowned theologian and keen writer, along the years, the Bishop published three books, the last one being "Più Nulla da Difendere", and wrote numerous works on "L'Osservatore Romano", being the only Italian bishop forming part of the Committee for the Catechism of the Catholic Church.
Confined to a wheelchair in his last years, during an homily, he announced to his priests and the people of the Diocese that he had lung cancer. He passed away on Tuesday, November 11, 2008, aged 77, after reciting the Rosary with his brother, whilst recovered at the Valduce Hospital of Como, around 22.30 pm., where he was admitted four weeks earlier. Till the end, until his health permitted, he served as confessor at Como's Duomo, including during the difficult periods trough which he received chemotherapy due to his malignant tumor.
His funeral Mass was celebrated at Como's Cathedral on Thursday, November 13, at 5 pm., presided by Cardinal Dionigi Tettamanzi, Archbishop of the Metropolitan See of Milan, with Bishop Coletti delivering the homily. The latter also read the spiritual testament of his predecessor, in which he asked to be buried in front of the altar of the Virgo Potens, inside the Cathedral of Como, inside a simple sarcophagus with the inscription: Alexander Maggiolini Ep. "Ne Mors Dissociet Quos Sociavit Amor". Originally interred inside the Cappella dei Canonici della Cattedrale at the Cimitero Monumentale di Como, his remains were transferred to cathedral and buried according to his wish, once a definitive tomb was finished.
