Born Andrew Holman Hamilton to Dr. Allen Hamilton and Helen (Knight) Hamilton, he spent the first half of his life in Fort Wayne, Indiana. He graduated from Williams College in Williamstown, MA and worked as a reporter for the Fort Wayne Journal-Gazette from 1932 to 1951. He married Suzanne Bowerfind in 1939. After serving as an officer in World War II, he returned to his journalist position and departed Fort Wayne in 1951 to pursue graduate studies at the University of Kentucky. He received a Ph.D. in 1954. He continued to serve on the faculty of the university until he retired in 1975.

He was the grandson of U.S. Congressman, Andrew Holman Hamilton (1834-1895). His cousins include physician and public health advocate Alice Hamilton, author Edith Hamilton, and actress Carole Lombard.
