James Patterson McClure was the son of Green McClure and Helen Patterson of Grayson County, Kentucky. His paternal grandparents were John Ashcraft McClure and Susannah Haycraft. His maternal grandparents were John Patterson and Rebecca Troutman. James was born in Grayson County on November 21, 1847. He was the husband of Harriet Lucretia Taylor, the daughter of Silas Taylor and second wife Nancy Kiper.

At the time of the 1880 Census of Leitchfield, Grayson County, Kentucky the James Patterson McClure family consisted of: J. P., a farmer, age 32; his wife Harriet K., age 29; and their children: Horace (Augustus), age 10, Nancy H. "Nannie", age 8; Jacob E(llis), age 6; and Alva D(ay), age 1. Also living with the family was his father, Green McClure, age 54; and his brother, J. F. McClure, age 22.

James P. McClure's wife, Harriet Taylor McClure, died on October 3, 1907. She was buried in the small family cemetery in the rear of #312 S. Clinton Street in the city of Leitchfield, Kentucky. James joined her there after his death on September 12, 1925. Also buried in this small family cemetery are their sons Alva Day McClure and Jacob Ellis McClure. Also buried there is Birdie Proctor McClure, who was the wife of both Alva and Jacob, and her daughter by Alva, Albirdie McClure.
