David W. Humphreys was commissioned Lieutenant of Company B, 2nd Mississippi, on March 4, 1861, and promoted to major on May 10. He was promoted again to lieutenant colonel on April 16, 1862.

Colonel Humphreys was killed at Gettysburg, during Pickett's Charge, on July 3, 1863. The extent of his wounds is unknown at this time.If he had advanced beyond the Emmitsburg Road, be would have fallen in the field between the road and the general area south of the Bryan Barn.

Col. Humphries was buried in an unmarked grave on the battlefield. His remains were probably removed in the 1870s to Hollywood Cemetery in Richmond, Virginia, and interred in the "Gettysburg Dead" section.
