He worked within the Vatican Diplomacy between 1889 and 1904, after passing with distinction the oral exam of the Concorso Diplomatico, serving among others as secretary of the Apostolic Nunciature in France, on personal request of the Apostolic Nuncio, Cardinal Domenico Ferrata, and later Auditor of the Apostolic Nunciature in Monaco.

Furthermore, Peri–Morosini, served among others as member of the Extraordinary Commission of the Holy See for the jubilee feasts of King Albert of Saxony, auditor of the Apostolic Nunciature in Brussels, under the Apostolic Nuncios Rinaldini and Granito Pignatelli di Belmonte, member of the Extraordinary Commission to Great Britain, following the demise of Queen Victoria and the Coronation of King Edward VII, auditor first class to Cardinal Rinaldini in the Apostolic Nunciature of Madrid, Spain and counselor of the Pontifical Embassy of King Alfonso XIII, of Spain.

Returning to Rome in 1902, he was appointed member of the Congregazione degli Affari Ecclesiastici Straordinari and assisted the Sacristan of His Holiness, Msgr. Pifferi whilst administering the last rites to the convalesent Pope Leo XIII.

Monsignor Peri–Morosini received his episcopal consecration with the Titular See of Arca in Phoenicia on April 24, from Rafael Cardinal Merry del Val y Zulueta inside the Basilica of the SS. Trinità dei Monti in Rome. He took possession of his see on April 24. Pope Benedict XV accepted the resignation of his see on December 29, 1916, which he abandoned following a diocesan crisis and was in turn named consulter within the Congregazione degli Affari Ecclesiastici Straordinari. The Prelate died aged 69, at Solcio di Lesa, upon the Lago Maggiore, on July 27, 1931. His remains where interred inside the communal cemetery of Lugano. They where later transferred to the chapel of canons in the same cemetery in 1948, and where later re-interred inside the crypt of the Sanctuary of the Sacred Heart of Jesus at Lugano, beside his predecessors in 1971.

During his episcopate, Bishop Alfredo contributed among others to further religious education in schools, promulgating new diocesan catechism, took care of the cathedral's restoration, the celebrations of the First Diocesan Synod and three diocesan Eucharistic Congresses.
